BONE MARROW PLASMA COLLECTION AND PROCESSING STANDARD OPERATING PROCEDURE (SOP) FOR SAMPLES IN BONE MARROW PLASMA DATASETS
- All participants will be fasting for over 6 hours at the time of bone marrow sampling as they underwent sedated bone marrow aspiration procedures and as such this reduces the interference or variability of recent dietary intake on metabolite levels.
- During the procedure, collect fresh bone marrow aspirate sample into “purple top” EDTA tubes that are on ice.
- Rapidly transfer bone marrow aspirate sample in EDTA tubes while on ice to processing laboratory.
- Pool bone marrow aspirate sample from EDTA tubes into a 50 mL conical tube.
- Spin the pooled EDTA tube(s) at 500xg for 10 minutes.
- Carefully remove pooled EDTA plasma and place into 15 mL conical tube.
- Spin the plasma at 500xg for 10 minutes.
- Aliquot the EDTA BM plasma into 2 mL Nunc vials and store at -80C.
- Record volume of aliquots on bone marrow processing worksheet.
- Label vials and the processing worksheet with appropriate bar coded label(s)
- Store aliquots in -80o freezer and record box and position location on processing worksheet.
